Which stat test do i need?

I need to compare sampling frequency of water quality data at a site to see if biweekly sampling is necessary or not. I am comparing biweekly to quarterly sampling over a period of 10 years. My instinct is to run a 2sample t-test and compare the means but is my data independent since my quarterly dates are also biweekly dates? Sample size for biweekly is n=255; sample size for quarterly is n=42. Any advice, hints?
