  • The influenza virus is known for its ability to cause acute febrile respiratory illness with possibility of severe morbidity and mortality in humans. Host immune response contributes to the disease in addition to the damage resulted from invasion of the virus. Cytokine storm with activation of the innate and the antigen specific acquired immunity is a major concern. In our transgenic mouse model system, influenza hemagglutinin (HA)-specific T cells respond to influenza virus infection and produce effector cytokines with both facilitated viral clearance and aggravated lung inflammation. Adoptive transfer of HA-specific naïve CD8+ T cells causes mortality with sub-lethal dose of influenza virus infection in mice. Co-transfer of HA-specific naïve CD4+ T cells impedes this lethal effect. The HA-specific CD4+ T cells have regulated the activity of effector CD8+ T cells. Balanced effector and regulatory arms of antigen-specific CD4+ T cells will provide efficient virus eradication without undesired tissue damage by the immune response. We will seek to dissect the molecular mechanisms responsible for maintenance of such an optimized balance. Identification of key molecules for the balance will offer diagnostic and therapeutic applications for severe influenza illness in humans.
